An HPLC Method to Determine Phenolic Compounds of Plant Extracts: Application to Byrsonima crassifolia and Senna alata Leaves

Objectives: method separating, identifying, quantifying mixture nine phenolic compounds (gallic acid, 3-hydroxybenzoic p-coumaric catechin, myricetin, rutin, quercetin, kaempferol, cyanidin) was developed, validated, applied to analyze aqueous hydroethanolic extracts from Byrsonima crassifolia (L.) Kunth Senna alata leaves. Materials Methods: separation carried out by HPLC, using Shim-pack VP-ODS C18 column (5 μm, 150 x 4.6 mm) at 40°C. Detection performed 254 nm occurred in 35 min. Results: optimized validated each compounds. calibration curve compound standards showed suitable linear fitting exhibited correlation coefficients greater than 0.990. LOD LOQ varied between 6.2807 - 14.8851 μg mL-1 6.8002 16.0071 mL-1, respectively. found be robust changes ±2 ml mobile phase composition. indicated contents gallic cyanidin whereas one did not show first substance. extract presented only acid rutin cyanidin, kaempferol were also identified one. Conclusion: HPLC is efficient, precise, accurate, sensitive determining plant it recommended efficient assays routine work.
